Research progression on molecular mechanisms of hepatoblastoma in children

Abstract:Hepatoblastoma(HB)is the most common malignant liver tumor in children,predominantly affecting in-fants and young children aged under 5 years.With the adoption of multidisciplinary comprehensive treatment strategies,the survival rate of HB has been significantly improved;however,the prognosis of high-risk patients remains poor,and the adverse effects of traditional chemotherapy have become increasingly prominent.Current studies have demonstrated that the pathogenesis of HB is associated with dysregulation of signaling pathways including the Wnt/β-catenin pathway,PI3K/AKT/mTOR pathway,and LIN28B/RAN/AURKA pathway,as well as abnormal activation of proto-oncogenes such as CTNNB1,C-myc,and N-myc.Additionally,abnormalities of various enzymes(e.g.,Polo-like kinase 1,Gankyrin)and aberrant RNA editing are also involved in the development of HB.In-depth investigation of the molecular mechanisms un-derlying HB will provide crucial theoretical support for the development of novel targeted therapeutic regimens and the im-plementation of precision medicine.
