Effect and mechanism of intraperitoneal injection of seaweed polysaccharide combined with budesonide nasal spray on allergic rhinitis rats

Abstract:Objective To observe the therapeutic effect of intraperitoneal injection of seaweed polysaccharide com-bined with budesonide nasal spray on allergic rhinitis(AR)rats,and to explore its possible mechanism.Methods Twen-ty-five SPF healthy SD rats were randomly divided into the normal control group,model control group,budesonide group,seaweed polysaccharide group,seaweed polysaccharide+budesonide group,with 5 mice in each group.In addition to the normal control group,the AR models were established by intraperitoneal injection of ovalbumin(OVA)as a sensitizing agent and nasal instillation of OVA nasal stimulant.After modeling was successfully completed,the budesonide group re-ceived nasal spray administration of budesonide,while the seaweed polysaccharide group received intraperitoneal injection of 200 mg/kg seaweed polysaccharide.The seaweed polysaccharide+budesonide group received both nasal spray and in-traperitoneal injection.The normal control group and model control group received the equal volume of normal saline solu-tion via nasal spray and intraperitoneal injection.After 2 weeks of treatment,the nasal symptoms and signs were observed and scored in each group.ELISA was used to detect serum interleukin-33(IL-33)and growth-stimulated expression gene 2(ST2),and real-time fluorescent quantitative PCR was used to detect IL-33 and ST2 mRNA expression in the nasal mu-cosa tissues.Results Rats in the normal control group did not have allergic rhinitis symptoms such as sneezing,runny nose,etc.,and occasionally scratched their noses;rats in the model control group successively showed the phenomenon of nose scratching and sneezing,and finally showed obvious nose scratching,sneezing and clear runny nose after stimula-tion;rats in the budesonide group,seaweed polysaccharide group,and seaweed polysaccharide+budesonide group had fewer nose scratches,sneezing and runny nose than rats in the model control group.Compared with the normal control group,the scores of nasal symptoms and signs,serum IL-33 and ST2 levels,and the relative expression levels of IL-33 and ST2 mRNA in nasal mucosa all increased in the model control group(all P<0.05).Compared with the model control group,the scores of nasal symptoms and signs,serum IL-33 and ST2 levels,and the relative expression of IL-33 and ST2 mRNA in nasal mucosa decreased in the budesonide group,seaweed polysaccharide group and seaweed polysaccharide+budesonide group,and the decrease was more obvious in the seaweed polysaccharide+budesonide group(all P<0.05).Conclusion The combination of seaweed polysaccharide intraperitoneal injection and budesonide nasal spray may re-duce the nasal symptoms of AR rats by reducing the level of IL-33 and ST2 in serum and nasal mucosa.
