Proliferation,migration,and invasion of laryngeal cancer cells with reduced expression of transcription factor FOXM1 and the activity of JAK2/STAT3 pathway

Abstract:Objective To observe the proliferation,migration,and invasion capabilities of laryngeal cancer cells with down-regulated transcription factor FOXM1 expression and the activity of the JAK2/STAT3 pathway,providing a ref-erence for targeted therapy of laryngeal cancer.Methods Human laryngeal cancer cells TU686 were divided into the control group,FOXM1 down-regulation group 1,and FOXM1 down-regulation group 2,which were transfected with nega-tive control siRNA,siRNA 1 targeting FOXM1,and siRNA 2,respectively.Transfection efficiency was verified using Western blotting and qRT-PCR.Cell proliferation was assessed using the CCK-8 assay at 24,48,and 72 h after transfec-tion.Cell migration and invasion capabilities were evaluated using the Transwell chamber assay.Western blotting was em-ployed to detect JAK2/STAT3 signaling pathway-related proteins,including JAK2,phosphorylated JAK2(p-JAK2),STAT3,and p-STAT3.Results At 24,48,and 72 of cultivation,the proliferation capacities of cells in the FOXM1 down-regulation group 1 and FOXM1 down-regulation group 2 were significantly lower than those of the control group(all P<0.05).The number of migrating and invading cells in the FOXM1 down-regulation group 1 and FOXM1 down-regulation group 2 was also significantly smaller than that of the control group(both P<0.05).The expression levels of p-JAK2 and p-STAT3 proteins in the FOXM1 down-regulation group 1 and FOXM1 down-regulation group 2 were lower than those in the control group(all P<0.05).There were no statistically significant differences in the expression levels of JAK2 or STAT3 proteins among the groups(all P>0.05).Conclusion FOXM1 down-regulation inhibits the proliferation,migration,and invasion capabilities of laryngeal cancer cells,and their mechanism may be related to the regulation of the phosphoryla-tion level of JAK2/STAT3 signaling pathway.
