Relationship between SNPs at rs11078928,rs2305479,and rs1031458 loci of GSDMB gene and asthma susceptibility in Zhuang children in Guangxi

Abstract:Objective To investigate the relationship between single nucleotide polymorphisms(SNPs)at the rs11078928,rs2305479,and rs1031458 loci of the gasdermin B(GSDMB)gene and asthma susceptibility in Zhuang chil-dren in Guangxi,and to provide a basis for personalized treatment of asthmatic children in this region.Methods Totally 118 Zhuang children with asthma were selected as the asthma case group,and 121 healthy Zhuang children who underwent physical examinations during the same period were selected as the healthy control group.Using polymerase chain reaction and high-throughput sequencing technology to detect the SNPs at the rs11078928,rs2305479,and rs1031458 loci of the GSDMB gene,and to calculate the genotype distribution frequency and allele distribution frequency at these loci.After ad-justing for age and gender,a binary Logistic regression analysis was used to investigate the associations between the GSD-MB gene variants rs11078928,rs2305479,and rs1031458 and the incidence of childhood asthma under four models:co-dominance,dominance,recessiveness,and over-dominance.We used the SHEsis software to perform linkage disequilibri-um and haplotype analysis between SNP loci,and analyzed the relationship between these haplotypes and the incidence of asthma in Guangxi Zhuang children.Results Compared with the healthy control group,the genotype distribution fre-quencies of the rs11078928,rs2305479,and rs1031458 loci in the GSDMB gene in the asthma case group were not signifi-cantly different(all P>0.05).In the asthma case group,the T allele accounted for 78.8%and the C allele for 21.2%at the rs11078928 locus of the GSDMB gene,while in the healthy control group,the T allele accounted for 69.0%and the C allele for 31.0%;a statistically significant difference was found between the two groups(both P<0.05).Similarly,at the rs2305479 locus of the GSDMB gene,the C allele accounted for 78.4%and the T allele for 21.6%in the asthma case group,compared with 69.0%for the C allele and 31.0%for the T allele in the healthy control group,and there was a sta-tistically significant difference(both P<0.05).Lastly,at the rs1031458 locus of the GSDMB gene,the T allele accounted for 78.4%and the G allele for 21.6%in the asthma case group,while in the healthy control group,the T allele accounted for 69.8%and the G allele for 30.2%,also showing a statistically significant difference(both P<0.05).At the rs11078928 locus of the GSDMB gene,under the co-dominant model,the TT genotype increased the risk of asthma in Guangxi Zhuang children in comparison with the CC genotype(OR=3.046,95%CI:1.006-9.217,P=0.049).Under the dominant model,the TT genotype also increased the risk of asthma in Guangxi Zhuang children as compared with the TC+CC genotype(OR=1.914,95%CI:1.130-3.242,P=0.016).At the rs2305479 locus of the GSDMB gene,un-der the dominant model,the CC genotype increased the risk of asthma in Guangxi Zhuang children as compared with the CT+TT genotype(OR=1.914,95%CI:1.130-3.242,P=0.016).At the rs1031458 locus of the GSDMB gene,under the dominant model,the TT genotype increased the risk of asthma in Guangxi Zhuang children as compared with the GT+GG genotype(OR=1.796,95%CI:1.060-3.042,P=0.030).There was a strong linkage disequilibrium between each pair of the rs11078928,rs2305479,and rs1031458 loci of the GSDMB gene.These loci formed two haplotypes,namely CTG(OR=0.618,95%CI:0.408-0.937,P=0.023)and TCT(OR=1.617,95%CI:1.067-2.452,P=0.023).Conclusions The SNPs at the rs11078928,rs2305479,and rs1031458 loci of the GSDMB gene are associated with asth-ma susceptibility in Guangxi Zhuang children.The haplotype CTG reduces the risk of asthma,while the haplotype TCT increases the risk of asthma.
