Effect of concentrated growth factor combined with Bio-Oss bone meal in repairing rabbit skull defect

Abstract:Objective To observe the osteogenic effect of concentrated growth factors(CGFs)combined with Bio-Oss bone meal in the early repair of rabbit skull defects.Methods Nine New Zealand male white rabbits were selected.We established the bone defect models with diameter of 15 mm on the cranial parietal bone by the right side of sagittal su-ture,and then randomly divided into three group(with 3 in each group).In the experimental group,the bone defect was filled with CGFs+Bio-Oss bone meal at a ratio of 1:1.The bone defect in the control group was only filled with Bio-Oss bone meal.The wounds of both groups above were covered with Bio-Gide film.No process was done on the blank group.Specimens were obtained at 6 weeks after surgery to observe the early repair of bone defects by cone beam CT(CBCT)im-ages,Hematoxylin and eosin(HE)staining,Masson staining,and Immunohistochemistry(IHC)staining.Results No obvious new bone formation was observed in the blank group by CBCT scan analysis.The control group exhibited increased new bone formation as compared with the blank group,and most of the new bone grew"centripetally"at the edge of bone defect.In the experimental group,a large number of new bones were formed both at the edge and the center of the bone de-fect and the gray value measurement analysis showed that the new bone mineral density of experimental group was signifi-cantly higher than that of the other groups,with statistically significant difference(P<0.05).HE and Masson staining showed that the repair effect of the experimental group was better than that of the other two groups,and the percentage of new bone area and the ratio of collagen fiber area of the experimental group were significantly higher than those of the other two groups(all P<0.05).The results of IHC staining showed that the positive expression levels of alkaline phosphatase(ALP)and osteocalcin(OCN)in the experimental group were significantly higher than those of the other two groups(all P<0.05).Conclusion In the early stage of bone repair,the combination of CGFs and Bio-Oss bone meal can achieve bet-ter osteogenic effect and reduce the infiltration of macrophages and other inflammatory factors.
