Relationships between rs1800469 and rs4803455 polymorphisms of TGF-β1 gene and susceptibility and prognosis of hepatocellular carcinoma

Abstract:Objective To investigate the relationships between rs1800469 and rs4803455 polymorphisms in trans-forming growth factor β1(TGF-β1)gene and genetic susceptibility and survival prognosis of hepatocellular carcinoma(HCC).Methods A total of 202 HCC patients(HCC group)and 176 healthy controls(control group)were selected.The single nucleotide polymorphisms(SNPs)of rs1800469 and rs4803455 loci of TGF-β1 gene were detected by mass spec-trometry.Telephone follow-up was conducted,and the last follow-up was up to December 2015,and death and survival were recorded.The goodness-of-fit χ2 test was used to analyze the Hardy-Weinberg genetic equilibrium of genotype distri-bution,the effects of rs1800469 and rs4803455 SNPs of TGF-β1 gene on the genetic susceptibility of HCC were analyzed by unconditional Logistic regression,and the relationships between rs1800469 and rs4803455 SNPs of TGF-β1 gene and the prognosis of HCC patients were analyzed by Cox proportional hazards model.Results The results of Hardy-Weinberg genetic equilibrium test showed that there was no significant difference in the frequency of each genotype between the two groups(P>0.05).Compared with the rs1800469 AA genotype,the risk of HCC in individuals with GG and GA+GG geno-types was lower(all P<0.05),and the risk of HCC in individuals with GA genotype was lower,but the difference was not statistically significant(P>0.05).Compared with the rs480345 CC genotype,the risk of HCC in individuals with CA,AA and CA+AA genotypes was lower,but the difference was not statistically significant(all P>0.05).Gender and HBV infec-tion were used as stratified variables.In the female population,compared with the rs1800469 AA genotype,the risk of HCC in individuals with GA+GG genotype was lower(P<0.05),and compared with the rs4803455 CC genotype,the risk of HCC in individuals with CA+AA genotype was lower(both P<0.05).In the male population,there was no significant difference in the risk of HCC among individuals with different genotypes of rs1800469 and rs4803455(all P<0.05),and HBV infec-tion had no effect on the risk of HCC in individuals with different genotypes of rs1800469 and rs4803455(P>0.05).Of the 202 patients,106 died and 96 survived.There was no significant difference in the survival time of each genotype at rs1800469 and rs4803455(all P>0.05).The SNPs of rs1800469 and rs4803455 of TGF-β1 gene had no effect on the prog-nosis of HCC patients(all P>0.05).Conclusions Individuals with TGF-β1 gene rs1800469 GG and GA+GG genotypes had a low risk of HCC.In the female population,individuals with the GA+GG genotype had a low risk of HCC;the SNPs of rs1800469 and rs4803455 were not found to be associated with the prognosis of HCC patients.
