Expression of LncRNA PINK1-AS and miR-134-5p in gastric cancer tissues and their relationships with clinicopathological parameters and prognosis

Abstract：Objective To investigate the expression of long non-coding ribonucleic acid(LncRNA)phosphatase and tensin homolog induced kinase 1 antisense ribonucleic acid(PINK1-AS),and microribonucleic acid-134-5p(miR-134-5p)in the gastric cancer tissues,and their relationships with clinicopathological parameters and prognosis.Methods Totally 104 gastric cancer patients who underwent surgical resection were selected,and RT-qPCR was used to detect the expression of LncRNA PINK1-AS and miR-134-5p in the cancer tissues and corresponding adjacent tissues,and we analyzed the relationships between LncRNA PINK1-AS,miR-134-5p expression and clinicopathological parameters in gastric cancer tissues.The correlation between the expression of LncRNA PINK1-AS and miR-134-5p in gastric cancer tis-sues was analyzed by Pearson method.Gastric cancer tissues were divided into high/low LncRNA PINK1-AS and miR-134-5p expression groups according to their median LncRNA PINK1-AS and miR-134-5p expression,and various survival curves were plotted using the Kaplan-Meier method.The mortality factors of gastric cancer patients were analyzed by Cox regression.Results The expression of LncRNA PINK1-AS in the gastric cancer tissues was higher than that in the adja-cent tissues,while the expression of miR-134-5p was lower than that in the adjacent tissues(both P<0.05).The expres-sion of LncRNA PINK1-AS and miR-134-5p in gastric cancer tissues was negatively correlated(r=-0.707,P<0.05).There were statistically significant differences in the expression levels of LncRNA PINK1-AS and miR-134-5p in gastric cancer tissues with different degrees of differentiation,TNM staging,and lymph node metastasis(all P<0.05).At 3-year follow-up,the overall survival rate of 104 gastric cancer patients was 61.54%(64/104).The 3-year overall survival rate was lower in the high LncRNA PINK1-AS expression group than that in the low LncRNA PINK1-AS expression group,and the 3-year overall survival rate was higher in the high miR-134-5p expression group than that in the low miR-134-5p expres-sion group(P<0.05).Poor differentiation,TNM stage Ⅲ,lymph node metastasis,and LncRNA PINK1-AS≥2.15 were independent risk factors for death in gastric cancer patients,and miR-134-5p≥0.64 was an independent protective factor(all P<0.05).Conclusion High expression of LncRNA PINK1-AS and low expression of miR-134-5p in gastric cancer tissues were associated with decreased differentiation,increased TNM stage,lymph node metastasis and poor prognosis.
