Effect of down-regulated CENPM gene expression on proliferation,apoptosis,invasion,and EMT phe-notype changes of prostate cancer PC3 cells

Abstract:Objective To observe the effects of down-regulated centromere protein M(CENPM)gene expression on the proliferation,apoptosis,invasion ability,and epithelial-mesenchymal transition(EMT)phenotype of prostate cancer PC3 cells.Methods Human prostate cancer PC3 cells were cultured in vitro and divided into the experimental group and control group.The experimental group was transfected with CENPM shRNA,while the control group was transfected with negative control shRNA.The cell proliferation ability was measured using the CCK-8 assay,and the proliferation rate was calculated.Apoptosis was detected using flow cytometry,and the apoptosis rate was calculated.The invasion ability of the cells was assessed using Transwell assay.EMT-related proteins(E-cadherin,Vimentin,and N-cadherin)were detect-ed by Western blotting.Results The experimental group showed lower cell proliferation rate,invasion ability,and expres-sion levels of Vimentin and N-cadherin proteins as compared with the control group;in contrast,the apoptosis rate and E-cad-herin protein expression were higher in the experimental group than in the control group(all P<0.05).Conclusion Down-regulation of CENPM gene expression can reduce the proliferation and invasion of prostate cancer PC3 cells,inhibit EMT,and promote apoptosis.
