Expression changes and clinical significance of ACOX1 and DUSP14 in colorectal cancer tissues

Abstract:Objective To investigate the expression changes of acyl-CoA oxidase 1(ACOX1)and bispecific phospha-tase 14(DUSP14)in the colorectal cancer(CRC)tissues and their clinical significance.Methods Ninety-eight CRC pa-tients who underwent surgical treatment were selected,and the cancer and adjacent tissues(more than 2 cm away from the tumor edge)were taken during surgery.Immunohistochemistry was used to detect the expression of ACOX1 and DUSP14 proteins in the tissues.Spearman rank correlation analysis was used to analyze the correlation between the expression of ACOX1 and DUSP14 proteins in CRC cancer tissues,as well as the relationship between their expression levels and the clini-cal pathological parameters;the patients were followed up,and we recorded their survival status,and used the Cox regression model to analyze the influencing factors for the prognosis of CRC.Results The positive expression rates of ACOX1 protein in the CRC cancer tissues and adjacent cancer tissues were 24.49%(24/98)and 84.69%(83/98),respectively,and the difference was statistically significant(P<0.05).The positive expression rates of DUSP14 protein in the CRC cancer tissues and adjacent cancer tissues were 79.59%(78/98)and 12.24%(12/98),respectively,and the difference was statistically significant(P<0.05).The expression of ACOX1 and DUSP14 proteins in CRC cancer tissues was negatively correlated(rs=-0.792,P<0.05).Compared with the CRC tissues at TNM stages Ⅰ and Ⅱ,with high and middle differentiation,and with-out lymph node metastasis,the positive expression rates of ACOX1 protein in the CRC tissues at TNM stage Ⅲ,with low de-gree of differentiation,and with lymph node metastasis were lower,and the positive expression rates of DUSP14 protein were higher(all P<0.05).During the follow-up period of CRC patients,42 cases died,and the 3-year overall survival rate was 57.14%(56/98).The 3-year survival rates of patients with positive and negative expression of ACOX1 protein were 87.50%(21/24)and 47.30%(35/74),respectively,with statistically significant differences between the groups(P<0.05).The 3-year survival rates of patients with positive and negative expression of DUSP14 protein were 48.72%(38/78)and 90.00%(18/20),respectively,with statistically significant differences between the groups(P<0.05).The negative expression of ACOX1 protein,the positive expression of DUSP14 protein,the TNM stage Ⅲ and lymph node metastasis were risk factors for the prognosis of CRC patients(all P<0.05).Conclusion In CRC,ACOX1 is low expressed and DUSP14 is highly expressed,and the expression changes of the two are related to tumor development and prognosis.
