Relationships between the expression of SEC62 and LSM12 and clinicopathological parameters,EMT,and prognosis in colorectal cancer tissues

Abstract:Objective To analyze the relationships between the expression of pre-protein translocation factor(SEC62)and RNA splicing factor(LSM12)and clinicopathological parameters,epithelial-mesenchymal transition(EMT),and prognosis in colorectal cancer(CRC)tissues.Methods Ninety-two CRC patients were selected,and CRC tissues and adjacent tissues were collected during surgery.Real-time fluorescence quantitative PCR was used to detect SEC62,LSM12 mRNA,and EMT-related genes(E-cadherin/N-cadherin,Twist transcription factor[TWIST]genes),and immunohistochemistry was used to detect SEC62 and LSM12 proteins.The relationships between the expression of SEC62 and LSM12 mRNA and clinical pathological parameters and EMT-related genes in the colorectal cancer tissues were analyzed.RNAseq data of cancer tissues from 643 CRC patients were downloaded from the Cancer Genome Atlas da-tabase(TCGA, the relationship between SEC62 and LSM12 mRNA expression and 5-year overall survival rate of CRC patients was analyzed.Results Compared with the adjacent tissues,the mRNA expression levels of SEC62,LSM12,N-cadherin,and TWIST were higher,the mRNA expression of E-cadherin was low-er,and the positive expression rates of SEC62 and LSM12 proteins were higher in the CRC tissues,with statistically signif-icant differences(all P<0.05).There were statistically significant differences in the expression levels of SEC62 and LSM12 mRNA in CRC tissues with different TNM stages,differentiation levels,and with or without lymph node metastasis(all P<0.05).The expression of SEC62 mRNA in CRC tissues was positively correlated with N-cadherin and TWIST mRNA expression(r=0.643,0.721,respectively;all P<0.05),and was negatively correlated with E-cadherin mRNA expression(r=-0.684,P<0.05).The expression of LSM12 mRNA in the CRC tissues was positively correlated with N-cadherin and TWIST mRNA expression(r=0.715,0.786,respectively;all P<0.05),and was negatively correlated with E-cadherin mRNA expression(r=-0.687,P<0.05).Based on the median expression of SEC62 and LSM12 mRNA,643 CRC patients in TCGA were divided into the high SEC62 mRNA expression group(>3.12,n=322),low expression group(≤3.12,n=321),high LSM12 mRNA expression group(>2.57,n=321),and low expression group(≤2.57,n=322),respectively.The 5-year overall survival rate of individuals with high expression of SEC62 mRNA was lower than that of individuals with low expression of SEC62 mRNA(Log-rank χ2=5.960,HR=1.547,95%CI=1.225-1.820,P<0.05);the 5-year overall survival rate of individuals with high expression of LSM12 mRNA was lower than that of individuals with low expression of LSM12 mRNA(Log-rank χ2=6.012,HR=1.649,95%CI=1.253-1.834,P<0.05).Conclusion SEC62 and LSM12 are highly expressed in the CRC tissues,and their expression changes are related to tumor TNM staging,differentiation,lymph node metastasis,EMT,and prognosis.
