Expression changes of miRNA-125a, STAT3, and IL-10 in cervical carcinoma tissues

Abstract:Objective To investigate the expression changes and significance of microRNA-125a (miR-125a),signal transduction and activator of transcription 3 (STAT3),and interleukin-10 (IL-10) in the cervical cancer tissues.Methods RT-PCR and qRT-PCR were used to detect the expression levels of miR-125a,signal transducer and activator of transcription 3 (STAT3),and IL-10 mRNA in 56 cases of cervical squamous cell carcinoma tissues (cervical cancer group) and 40 cases of normal cervical epithelial tissues (control group),and we analyzed the correlations between them in the cervical cancer group.The immunohistochemical Elivision two-step was used to detect the protein expression of STAT3 and IL-10 in the two groups.The relationship between the expression of miR-125a,STAT3 mRNA and clinicopathological features of cervical cancer patients was analyzed.Results Compared with the control group,the expression of miR-125a mRNA decreased in the cervical cancer group,and the mRNA expression of STAT3 and IL-10 increased (all P <0.05).The expression levels of STAT3 and IL-10 were positively correlated (r =0.457 8,P < 0.05).Compared with the control group,the positive expression rate of STAT3 protein in the cervical cancer group increased (P < 0.05).The expression of miR-125a was related to the differentiation of cervical cancer,lymph node metastasis,and clinical stage (all P < 0.05).The expression of STAT3 was related to lymph node metastasis and clinical stage (both P < 0.05).Conclusion The expression of miR-125a decreases,and the expression of STAT3 and IL-10 increases in the cervical cancer tissues.The three factors may interact and participate in the occurrence and development of cervical cancer.
