Effect of regulating expression of small molecule GTP binding protein Rab27 on Cyclin, p-FAK and p-IκB expression of human bladder cancer cells

Abstract:Objective To observe the effect of regulating the expression of small molecule GTP binding protein Rab27 on the cell cycle protein (Cyclin),phosphorylated focal adhesion kinase antibody (p-FAK) and nuclear factor κB (pIκB) of human bladder cancer cells.Methods Rab27 plasmid and Rab27 siRNA were transfected into BIU-87 and 5637 cells in logarithmic phase,respectively.The levels of Rab27,Cyclin D1,Cyclin E,p-FAK (Tyr925),p-FAK (Tyr576/ 577) and p-IκB were analyzed by Western blotting in the bladder cancer BIU-87 and 5637 cells before and after transfection.Results Compared with the Rab27 plasmid transfected BIU-87 cells before transfection,the relative expression of Rab27 was increased,and the expression of Cyclin D1,Cyclin E,p-FAK (Tyr925),p-FAK (Tyr576/577) and p-IκB was increased (all P < 0.05).After the Rab27 interference plasmid was transfected into 5637 cells,the relative expression of Rab27 was decreased (all P < 0.05),and Cyclin D1,Cyclin E,p-FAK (Tyr925),p-FAK (Tyr576/577) and p-IκB relative expression was decreased (all P <0.05).Conclusion After the Rab27 plasmid is transfected into BIU-87 cells,the expression of Cyclin D1,Cyclin E,p-FAK (Tyr925),p-FAK (Tyr576/577) and p-IκB is increased.After the Rab27 interference plasmid is transfected into 5637 cells,the expression of Cyclin D1,Cyclin E,p-FAK (Tyr925),p-FAK (Tyr576/577) and p-IκB is decreased.Rab27 may affect the cell cycle of bladder cancer by regulating the expression of NF-κB and FAK signaling pathway.
