Homology analysis and multidrug resistance mechanisms of carbapenem-resistant Acihetobacter baumanii

Abstract:Objective To evaluate the homology of resistance to carbapenems of Acinetobacter baumannii (AB)and its resistance mechanism to imipenem.Methods The molecular classification was conducted in 140 strains of AB from clinical collection by using amplified ribosomal DNA restriction enzyme analysis (ARDRA),enterobacteriaceae repeat gene conserved sequence-PCR (ERIC-PCR)is applied to do the homology analysis of drug resistance of 90 strains of AB.The outer membrane protein CarO and efflux pump gene AdeB mRNA expression was detected in 20 strains of resistant strains and 20 strains of sensitive strains by fluorescence quantitative PCR.Results Totally 130 strains were identified as AB by ARDRA in 140 strains.After ERIC-PCR genotyping,90 strains of AB were divided into 6 types,mainly for type A (65 strains),type B (19 strains),type C,D,F and E (which were scattered in the strains).The outer membrane protein CarO mRNA expression in the AB with drug-resistant strains was lower than that of the sensitive strains,while the efflux pump AdeB mRNA expression was higher (all P <0.05).Conclusions Type A and type B were mainly found in AB gen-otypes and there were scattered transmission of clone strains with resistance gene.The low expression of outer membrane protein Caro and high expression of efflux pump gene AdeB was the important mechanism of AB resistance to imipenem.
