Influence of Schisandrin B on proliferation of glioma U87 cells and its mechanism

Abstract:Objective To investigate the effect of Schisandrin B ( Sch B) on the proliferation of glioma U87 cells and to explore its possible mechanism. Methods Glioma U87 cells were cultured and were divided into groups A, B, C and D. Group D was added with culture solution that containing 5% serum 200 μL, and groups A, B and C were added with the same amount of nutrient solution which respectively contained 50, 100 and 200μg/ml Sch B. The cell proliferation was compared between these groups. The cell cycle and the expression of Caspase-3, Bax, Bcl-2 and Bax/Bcl-2 were respec-tively detected by ELISA and immunohistochemistry. Results Compared with group D, the A value at 48 h and 72 h in the groups A, B and C was decreased. Compared with groups D and A, the A values of Caspase-3, Bax and Bax/Bcl-2 and the positive expression rate of group B were increased (all P<0. 05). Compared with group D, the A values of Caspase-3 and Bax/Bcl-2, and the positive expression of group C were increased (all P<0. 05). Compared with group A, the A val-ue of Caspase-3 and the positive expression rate of group C were increased (all P<0. 05). Compared with group D, the cell percentage of the G1 and G2/M phases of group A was increased, all P<0. 05. Conclusion Sch B can significantly inhibit the proliferation of U87 cells, induce the apoptosis, and it has a dose-effect relationship, whose mechanism may be related with the up-regulation of Caspase-3, Bax and Bcl-2 expression.
