Effects of potato extracts on apoptosis and proliferation of non-small-cell lung cancer cells and its mechanism

Abstract:Objective To investigate the inhibitory effect of potato extracts on non-small-cell lung cancer ( NSCLC) A549 cells and the influence on apoptosis-related proteins Bax and Bcl-2.Methods The cultured A549 cells were divided into the control group and observation group .Different concentrations of potato extracts (5.0, 12.5, 25.0 and 50.0 μL) were added into the A549 cells of the observation group , and then the A549 cells were cultured for 24, 48 and 72 h (3 holes were set in each concentration ) .The control group was not added with any potato extract .The apoptosis was observed after 24 h of treatment by inverted fluorescence microscope .And the inhibition rate was obtained by OD value in 492nm by MTT method.The Bax and Bcl-2 mRNA expression was detected by RT-PCR.Results Under the fluorescence micro-scope, the cells were growing strong , the distribution was dense and the shape of the cells was spindle .The cells in the ob-servation group showed apoptosis at different time points .Meanwhile, the cell inhibition rate in the group with high concen-tration of potato extracts was higher (all P<0.05).The cell inhibition rate was also higher in the group with the same con-centration but longer intervention time (all P<0.05).Bax mRNA expression in each observation groups was higher than that of the control group.However, Bcl-2 expression was lower than that of the control group (all P<0.05).Conclusion Potato extracts can promote the apoptosis of A 549 cells and inhibit the cell proliferation , whose mechanism may be relat-ed to up-regulation of Bax and down-regulation of Bcl-2 expression .
