Influence of TLR3 activation on expression of reactive oxygen species in melanocytes

Abstract:Objective To investigate the effect of Toll-like receptor 3 (TLR3) activation on the expression of reactive oxygen species ( ROS) in melanocytes .Methods Primary melanocytes were isolated from normal human skin and cultured in vitro, then they were inoculated into six pore plates .They were divided into the experimental group and the control group with 9 holes in each group.The cells in the experimental group were added with TLR 3 ligand poly (I∶C) to inoculate for 2, 6 and 24 hours, while the cells in the control group was not added .The expression of ROS was measured by CM-H2DCFDA probe method combined with flow cytometry .ResultsWith the extension of incubation time , the expression of ROS in the melanocytes was increased in the control group , but there was no statistically significant difference .With the extension of incubation time , the expression of ROS in the melanocytes was significantly increased in the experimental group (all P<0.01).At each time point, the expression of ROS in the melanocytes of the experimental group was higher than that of the control group (all P<0.01).Conclusion The TLR3 activator poly(I∶C) could induce the increase of ROS in the melanocytes .
