Influence of secretion of human umbilical vein endothelial cells on their own migration and invasion

Abstract:Objective To observe the influence of the secretion of human umbilical vein endothelial cells (HuVECs) on their own migration and invasion .Methods The passage HuVECs were divided into 3 groups: groups A, B and C. Groups A and B were added with a certain concentration of FBS and HuVECs supernatant with 24-hour and 48-hour cultiva-tion, meanwhile, group C was added with a certain concentration of FBS and pure DMEM culture solution .Cell scratch as-say was used to observe cell migration distance .The number of cell migration and invasion of HUVECs was observed with Transwell.Results The migration distance of groups A, B and C was (219.27 ±28.44), (163.64 ±19.67) and (158.18 ±16.36) μm, respectively.Significant difference was found between group A and groups B and C ( all P<0.05);and no significant difference was found between group A and group B (P>0.05).The migrated cells in the group A were more than those of groups B and C (all P<0.01), and no significant difference in the migrated cells was found be-tween group B and group C (all P>0.05).The invasive cells of group A were less than those of group B , and the invasive cells of group B were less than those of group C (all P<0.01).Conclusion The secretion of HUVECs may promote the cell migration and inhibit the cell invasion to a certain extent .
