Changes in levels of serum inflammatory factors, expression of NF-κB in PBMC and immune cells in peripheral blood of patients with primary insomnia

Abstract:Objective To observe the changes in levels of the serum inflammatory cytokines interleukin ( IL)-2 and IL-6, nuclear factor (NF)-κB of peripheral blood mononuclear cell (PBMC) and immune cells in the peripheral blood of patients with primary insomnia, and to investigate the significance.Methods We detected the levels of IL-2, IL-6 and NF-κB of PBMC by ELISA and immune cells by hematology analyzer in 38 patients with primary insomnia ( observation group) and 42 cases of healthy check-up people (control group).The correlations among the indexes were detected by Spearman related analysis.Results In the observation group, IL-2 level was (97.78 ±30.61) ng/mL, IL-6 level was (6.05 ±2.01) pg/mL, PBMC NF-κB level was (9.06 ±5.74) pg/mL, leukocyte level was (7.01 ±3.43) ×109/L, lymphocyte level was (5.17 ±0.54) ×109/L and monocyte level was (0.60 ±0.21) ×109/L;in the control group, they were (77.82 ±35.85) ng/mL, ( 4.45 ±2.18) pg/mL, (4.21 ±2.29) pg/mL, (5.11 ±1.28) ×109/L, (4.92 ± 0.42) ×109/L and (0.44 ±0.21) ×109/L.Significant difference was found in the above indexes between the two groups (all P<0.05).In the observation group, the serum IL-2 was positively correlated with IL-6 (r=0.529, P<0.05), and was positively correlated with NF-κB (r=0.631, P<0.05), and IL-6 was positively correlated with NF-κB (r=0.809, P<0.05).Conclusions The serum levels of IL-2, IL-6 and PBMC NF-κB and immune cells are increased in patients with primary insomnia.The body immune function and inflammatory state may change when the human body suffers from chronic insomnia for a long time.
