Effect of 2-hydroxypropyl-β-cyclodextrin on migration and proliferation of HUVECs

Abstract:Objective To investigate the effect of 2-hydroxypropyl-β-cyclodextrin (2HP-β-CD) on the migration and proliferation of human umbilical vein endothelial cells ( HUVECs) in vitro.Methods HUVECs were cultured in vitro and were divided into two groups:the experimental group and the blank group .HUVECs in the experimental group were treated with different concentrations of 2HP-β-CD (10 -10-10 -5 mol/L), and the same amount of 0.5% FBS was added into the blank group .CCK-8 was used to detect the cell proliferation activities of the two groups , and the modified Boyden Chamber was applied to observe the cell migration in each group .Results The OD450 of the experimental group which was added with 10 -10 mol/L, 10 -9 mol/L and 10 -8 mol/L 2HP-β-CD was 1.4, 1.6 and 1.7 times higher than that of the blank group;and 10 -10 mol/L 2HP-β-CD had the most significant effect in promoting the proliferation of HUVECs ( all P <0.01).No significant difference was found in OD450 of the two groups when we added 10 -7, 10 -6 and 10 -5 mol/L 2HP-β-CD to HUVECs.The migration of HUVECs which were added with 2HP-β-CD at the concentration of 10 -9 mol/L and 10 -8 mol/L in the experimental group was significantly higher than that of the blank group (all P<0.05).When we added other concentrations of 2HP-β-CD, no significant difference was found in the cell migration between the experimental and blank groups.Conclusion Suitable concentrations of 2HP-β-CD can promote the migration and proliferation of HUVECs .
