Expression and significance of ASC and its subtype in PBMCs of patients with gouty arthritis

Abstract:Objective To observe the expression changes of apoptosis-associated speck-like protein containing a CARD (ASC) and its subtype (ASC-b) in the peripheral blood mononuclear cells (PBMCs) in patients with gouty arthritis ( GA) and to investigate the clinical significance .Methods The expression levels of ASC mRNA and protein and nuclear factor (NF)-κB p65 protein in the PBMCs were detected in 48 cases of patients with acute gouty arthritis (AGA group), 48 cases of patients with gouty arthritis in the intermission (IGA group) and 48 cases of healthy controls (HC group) by u-sing real-time fluorescent quantitative polymerase chain reaction ( PCR) and Western blotting .Results The expression of ASC mRNA in the AGA group was significantly higher than that of the IGA and HC groups (all P<0.01), and there was no difference between IGA and HC groups .The expression of ASC protein in the AGA and IGA groups was significantly lower than that of the HC group (all P<0.01), and the IGA group was the lowest (P<0.01).The expression of ASC-b protein in the AGA group was significantly higher than that of the IGA and HC groups (all P<0.01), and the IGA group was the lowest (P<0.05).The expression of NF-κB p65 protein in the AGA group was significantly higher than that of the IGA and HC groups (all P<0.01), and there was no statistical difference between the IGA and HC groups .Conclusion The expression of ASC and its subtype ( ASC-b) in the PBMCs of patients with AGA is abnormal , which indicates that ASC and ASC-b may play a critical role in the pathogenesis of GA .
