Effect of STAT3 on the proliferation and apoptosis of human intrahepatic cholangiocarcinoma cells

Abstract：Objective To investigate the effect and mechanism of signal transducer and activator of transcription 3 (STAT3) on the proliferation and apoptosis of human intrahepatic cholangiocarcinoma cells.Method We detected the STAT3 protein and mRNA levels in human cholangiocarcinoma cells by Western blotting and real-time PCR.We divided the cells of the intrahepatic group into the interference group and the control group 1, and cells were treated with the adenovirus 5μL for 48 h to knockdown the expression of STAT3.Furthermore, We divided the cells of the intrahepatic group into the overexpression group and the control group 2, and the cells were treated with the adenovirus 5μL for 48 h to over-express the expression of STAT3. We used the CCK8 to detect the cell proliferation of all group.We used the PI-AnnexinV to detect the cell apoptosis of all group. Results The mRNA and protein levels of STAT3 in the intrahepatic group were higher than those in the extrahepatic group (all P<0.05).The RBE and 9810 cell proliferation abilities of the interference group were lower than those of the control group 1(all P<0.05);and the RBE and 9810 cell apoptosis rates of the interference group were higher than those of the control group 1 (all P<0.05).The RBE and 9810 cell proliferation abilities of the overexpression group were higher than those of the control group 2 (all P<0.05);and the RBE and 9810 cell apoptosis rates of the overexpression group were lower than those of the control group 2 (all P<0.05).Conclusion STAT3 plays an important role in promoting intrahepatic cholangiocarcinoma cell prolifera-tion and anti-apoptosis.
