Inhibitory effect of crocetin on the TLR4/NF-κB pathway in RAW264 .7 macrophage and its mechanism

Abstract:Objective To investigate the inhibitory effect of crocetin on Toll-like receptor 4 ( TLR4)/NF-κB pathway in RAW264.7 cell line and its mechanism .Methods The murine macrophage RAW264.7 cell line was routinely cultured , MyD88 siRNA was designed and synthesized .RAW264.7 cell was respectively stimulated by LPS and treated with crocetin and MyD88 siRNA simultaneously .The expression of IL-6, TNF-α, iNOS, MCP-1 and IFN-βin the RAW264.7 macrophage was measured by real-time PCR, the protein expression of p65 and p-JNK1/2 was detected by Western blotting , and the ex-pression of IL-6 and TNF-αwas measured by ELISA .Results Crocetin decreased the mRNA expression of IL-6, TNF-α, iNOS and MCP-1, down-regulated the protein expression of p-JNK1/2 and p65, and also decreased the levels of IL-6 and TNF-α;MyD88 siRNA blocked the effect of crocetin on RAW264.7 cells stimulated by LPS.However, the blocking effect of crocetin and MyD88 siRNA on the IFN-βmRNA expression was not significant .Conclusion The inhibitory effect of crocetin on the TLR4/NF-κB pathway in RAW264.7 macrophage may be involved in MyD88 dependent pathway.
