Insulin resistance and adipocytokines changes in the Bama miniature pig with type 2 diabetes mellitus model

Abstract：Objective To establish Bama miniature pig with type 2 diabetes mellitus ( T2DM) model which induced by high fat and high sugar diet and intraperitoneal injection of streptozotocin (STZ), and to investigate the changes of insu-lin resistance and adipocytokines in the Bama miniature pig with type 2 diabetes mellitus model .Methods Firstly, 10 healthy male Bama miniature pigs were equally randomly divided into control group and diabetic group ( model group ) . Secondly , the pigs of control group was fed with normal diet , while the pigs of model group was fed with high fat and high carbohydrate.After feeding for 10 months, the model group was injected with STZ 100 mg/kg, the injection was repeated a week later, while the control group was injected with the same amount of sodium citrate solution .Finally, the observation of the laboratory targets including FPG , FINS, Leptin and Resistin were carried on .Meanwhile, the insulin resistance index ( HOMA-IR) was calculated .Results After the early stage of experiment , the levesl of weight , FPG, FINS, HOMA-IR increased significantly in the model group comparing with the control group (all P<0.01).After administration of STZ, the serum level of FPG increased significantly , and the level of FINS was lower in model group as compared with control group (all P<0.01).The levels of Leptin and Resistin were higher in the model group comparing with the control group (all P<0.05) and the correlation analysis showed that HOMA-IR was positively correlated with the levels of Leptin and Resistin in the model group (all P<0.05).Conclusion The levels of leptin and resistin are associated with the insulin resistance of T2DM miniature pig.
