Significance and expression of Gab1 and PI3KCA in bile duct adenocarcinoma tissue

Abstract：Objective To observe the expression of Gab1 and PI3KCA in human cholangioadenocarcinoma and dis-cuss the significances .Methods In 76 patients diagnosed as cholangioadenocarcinoma , 76 cancer tissues ( adenocarcino-ma group ) and 45 adjacent normal tissues ( adjacent cancer group ) were collected in operations .Immunohistochemical method, Western blotting and(qRT)-PCR were used to detect the expression of Gab 1 and PI3KCA in the two groups, and the relationship between the two genes and communication with clinicopathological indexes were discussed .Results Gab1 and PI3KCA expression in cholangiocarcinoma tissues were higher than those in normal bile duct tissues (all P<0.05). Gab1 and PI3KCA expressions were positively correlated with tumor diameter , TNM stage and lymph node migration ( all P<0.05).Spearman rank correlation analysis showed there was positive correlation between Gab 1 and PI3KCA(r=0.214, P<0.05).Conclusion Gab1 and PI3KCA are up-regulated in cholangioadenocarcinoma at cell , protein and mRNA lev-els, and may be used to predict the outcomes and become a future candidate for target therapy .
