Effect of ecdysterone on expression of insulin receptor substrate 2 in liver cell of type 2 diabetic rats

Abstract:Objective To investigate the effect of ecdysterone on the expression of insulin receptor substrate 2 ( IRS-2 ) in liver cell of type 2 diabetic rats .Methods 14 of the 42 rats were randomly selected to the normal control group , and was given ordinary rat food .The other 28 rats were made into type 2 diabetes mellitus rat models by using the high fatty diet and small dose of SIZ , and then diveided into model group and treatment group .The rats in model group or treatment group was given high fatty diet or high fatty diet fecdysterone .IRS-2 protein and mRNA in liver tissues of rats were detected by immunohistochemistry assay and RT-PCR assay after five weeks .Results The expression of IRS-2 protein and mRNA in normal group and treatment group were higher than those of the model group (P<0.01or <0.01).Conclusions Ecdyste-rone led to up-regulation expression of IRS-2 protein and IRS-2 mRNA of liver tissue in diabetic rats .This might play a role in ecdysterone-improved hepatic insulin resistance .
