Abstract:Objective To investigate the effect of mild hypothermia on the expression of P-glycorprotein ( P-gp) in primary cultured rat brain microvascular endothelial cells (RBMECs).Methods The primary brain microvascular endo-thelial cells of rats were divided into three groups:37 ℃group, 35 ℃group and 32 ℃group, and they were cultured for 5 days under the respective temperature .The immunoreactivity of P-gp was examined by immunocytochemistry and the ex-pression changes of P-gp in RBMECs were measured by Western blotting .Results Western blotting analysis showed that the expression level of P-gp in the 37 ℃group was higher than that of the other two groups (all P<0.01), and the protein expression level of the 32 ℃group decreased as compared with that of the 35 ℃ group (P<0.05).Conclusion Mild hypothermia may significantly affect the expression of the P-gp.Therefore , it has effect on the pharmacokinetics and phar-macodynamics .
