Abstract:Objective To investigate the effects of Hulan hypoglycemic sustained-release tablets (TGE) on glucose metabo-lism and hemorheology in rats with type 2 diabetes mellitus (T2DM).Methods The T2DM model was established by intraperitone-al injection of streptoptocin (STZ) 40 mg/kg.Then the Wister rats were randomly divided into 6 groups:metformin group, TGE high-dose, medium-dose, low-dose groups, model group and normal control group, which were respectively treated with metformin hydrochloride, TGE (1.2 g/kg), TGE (0.6 g/kg), TGE (0.3 g/kg) and normal saline for 4 weeks of continuous lavage.During the administration, the body mess was recorded every week as well as the amount of water and food intake everyday .After the experi-ment, the fasting blood glucose (FPG) and oral glucose tolerance (OGT) were determined. After the last administration, rats were fasted for 24 h, and the hemorheology indexes were measured. Results Compared with the model group, the FPG, the amount of water and food intake in the high-dose TGE and medium-dose TGE group were decreased (all P<0.01);the glucose and blood glu-cose curve declined after glucose load 0.5 and 2 h in the high-dose TGE and medium-dose TGE groups, at 0, 0.5 and 2 h in the met-formin group (all P<0.05 or P<0.01);all the indexes of hemorheology improved significantly (all P<0.05).Conclusion TGE at the dose of 0.6 and 1.2 g/kg has good regulating effect on glucose metabolism of rats with type 2 diabetic mellitus, and meanwhile improves the drinking water and food intake as well as the hemorheology indexes.
