Preparation and observement of heterotopic osteoinduction of β-TCP/rhBMP-2 composite
Abstract:To determine the osteoinductive potential of β-TCP/rhBMP-2 composite,the β-TCP/rhBMP-2 composite was obtained by combining Beta-tricalcium phosphate (β-TCP) prepared with recombinant human morphogenetic protein-2 (rhBMP-2)and was implanted into muscle pouches in the thigh of mice. β-TCPalone was implanted on the opposite site as controls.At intervals of 24 and 72 hours and 1,2,4 and 8 weeks after the implantation,the specimens were obtained,and histologic study and alkaline phosphatase assay(1,2,4 weeks)were performed.It was found that there was a large amount of new cartilage and bone formation within the composite,and incereasing with time.Meanwhile,there was no new bone formation where β-TCP alone was implanted.The levels of alkaline phosphatase in the β-TCP/rhBMP-2 implants also were increasing with time and were higher than those in controls (P<0.001).The results suggest that β-TCP/rhBMP-2 possesses heterotopic osteoinductive potential.
