Extraction and Purification Technologies of Flavonoids from Salvia plebeia R.Br.and Study on Its Antiviral Mechanism

Abstract:In this study,the ultrasound-assisted aqueous two-phase extraction combined with AB-8,HPD-100,D101 and NKA-9 macroporous resin purification was used to enrich the total flavonoids of Salvia plebeia R.Br.(SF),and the single factor and orthogonal tests were designed to screen optimal parameters of the aqueous two-phase extraction system and optimal purification process with macroporous resin.The results showed that 28%polyethylene glycol(PEG)1000,14%ammonium sulfate[(NH4)2SO4],0.4%NaCl,pH 3 and temperature of 40 ℃ were the optimum parameters for SF extraction.Through single factor tests,the best purification process was determined as static adsorption for 3 hours with 80 mg/mL sample solution at pH 3 and 10 BV of volume,and then elution with 60%ethanol solution at 6.0 BV.The results of pharmacodynamic experiments indicated that high dose[2.8 mg/(g·d)]SF could activate the immune activity by inhibiting the expression of α tumour necrosis factor(TNF-α),Toll-like receptor(TLR3 and TLR4),and increasing the ex-pression of interleukin 2(IL-2)and γ interferon(IFN-γ),and showed the significant anti-herpes simplex vi-rus Ⅰ type(HSV-Ⅰ)activity.
