Enzymatic Digestion Process Optimization of Cynanchum bungei Decne.Seed Protein and Study on Its Antioxidant Mechanism

Abstract:Using Cynanchum bungei Decne.seeds as experimental materials,this study optimized the en-zyme digestion process of seed protein through one-way tests and response surface analysis,and constructed a model for oxidative damage of human hepatocellular carcinoma cells HepG2 by using H2O2 to evaluate the pro-tective effect of the enzymatic digestion products against oxidative damage of the cells and preliminarily reveal the mechanism of antioxidant effects.The results showed that the optimal enzyme digestion conditions were as follows:flavored protease as hydrolase,liquid-solid ratio of 100∶1,enzyme addition of 8 100 U/g,and en-zyme digestion time of 3.1 h.The optimal purification reagent was 60%saturated ammonium sulfate.The<3 kDa(250 μg/mL)and 3 to 10 kDa(500 μg/mL)fractions were able to promote the transfer of Nrf2 protein to the nucleus,activate the expression of related antioxidant genes,regulate the synthesis of antioxidant en-zymes,and reduce oxidative damage.The results of this study could lay theoretical foundations for further re-search on C.bungei proteins and antioxidative peptide,and open up a new way for efficient utilization of C.bungei.
