Effect on Controlling Bacterial Leaf Spot of Ligustrum lucidum and Biological Characteristics of Bacteriophage

Abstract:Pseudomonas syringae is the main bacterium causing bacterial leaf spot of Ligustrum lucidum in China.Bacteriophage therapy is a potential biological control method at present with advantages of high safe-ty and low production cost.In this study,a P.syringae phage was isolated and purified from floral soil,and its morphological characteristics,biological characteristics and biocontrol efficacy were investigated.The results showed that a phage with strong lysis ability was obtained by using P.syringae as the host bacterium.The phage could form a transparent plaque.The titer of the phage was stable in the pH range of 4.0 to 10.0,and decreased significantly when the temperature was above 60 ℃.Its optimal multiplicity of infection was 0.1.The one-step growth curve showed that the incubation period was about 10 min,and the outbreak period was 130 min.It showed high tolerance to chloroform,but was sensitive to ultraviolet(UV)light.Antibacterial test showed that the bacterial count in leaves of L.lucidum was significantly lower in the phage treated group com-pared to the control group on the 4th day after phage inoculation.The phage could provide a useful exploration for the prevention and treatment of bacterial leaf disease caused by P.syringae.
