Inhibition of Fusarium oxysporum and Plant Growth Promotion by Bacillus amyloliquefaciens HX-23

Abstract:In this study,the antagonistic strains against Fusarium oxysporum were selected using the fil-ter paper disc method and the plate confrontation method,and were identified through morphological and mo-lecular biological characterization.Their biocontrol mechanisms were preliminarily investigated by assessing the inhibitory activities of cell-free filtrates and volatile organic compounds,as well as the abilities to produce ex-tracellular enzymes,growth-promoting substances,and biofilms.Additionally,the control efficacy against pep-per Fusarium wilt and the growth-promoting effects on seedlings were evaluated through in vitro leaf assays and pot experiments.The results indicated that a strain with stable inhibitory activity against F.oxysporum was screened out and designated as HX-23,showing an inhibition rate of 67.99%.It was identified as Bacillus amyloliquefaciens.The cell-free filtrate of strain HX-23 could effectively inhibit the growth of F.oxysporum,and at the concentration of 30%,the inhibition rate in mycelial growth and spore germination were 46.92%and 60.28%,respectively.Furthermore,it induced hyphal swelling,deformation and even rupture,resulting in the leakage of intracellular contents.The inhibition rate of F.oxysporum by the volatile organic compounds produced by strain HX-23 was 18.77%,which also led to hyphal deformity and reduced spore production.In addition,the strain demonstrated the ability to produce extracellular enzymes,siderophores,indole-3-acetic acid,along with capacities for phosphate solubilization and biofilm formation.In vitro leaf experiments showed that HX-23 strain could effectively control pepper Fusarium wilt,with a relative prevention and control effect of 23.16%.HX-23 strain could significantly promote the growth of pepper seedlings,increasing the fresh weight and dry weight of the whole plant by 17.70%and 22.35%,respectively.In conclusion,the strain HX-23 exhibited significantly antagonistic activity against F.oxysporum and growth-promotion effect on the plant,highlighting its potential as a biocontrol agent and providing a theoretical basis for its application.
