Cold Resistance Evaluation of Xinjiang Pear Germplasms Based on Electrolyte Leakage Method and Logistic Equation

Abstract:This study aimed to evaluate the cold resistance of 50 local pear germplasms from Xinjiang u-sing the electrolyte leakage method in conjunction with the Logistic equation for analysis.One-year-old dormant branches were selected as test materials and exposed to artificial low-temperature treatments at 4℃(control),-15,-18,-21,-24,-27,-30℃,and-33℃for 24 hours.The electrolyte leakage rate(ELR)was measured under different low-temperature treatment,and the changes of branch injury rate were calculated.The Logistic equation was applied to fit the data,and the semi-lethal temperature(LT50)was calculated for each germplasm.The results showed that the ELR of the pear germplasms was significantly or extremely signifi-cantly negatively correlated with the treating temperature.The ELR of 46 germplasms gradually increased as the temperature decreased,exhibiting an"S"shaped trend overall,and other 4 germplasms increased first and then decreased.Despite different pear germplasms showed consistent responses to low-temperature stress in general,the specific temperature points for their ELR significantly changing varied among germplasms.Under different low temperature,cellular injury percentage of the 50 pear germplasms increased in various magnitude.During the initial phase of temperature decreasing,the cell injury percentage in the majority of germplasms ex-hibited relatively minimal tend,but then it showed increasing trend overall with the treating temperature con-tinuously decreasing.The LT50 for all the 50 pear germplasms ranged from-20.20℃to-32.03℃.Through cluster analysis,the germplasms were classified into three groups based on cold resistance:Group Ⅱ,charac-terized by strong cold resistance,including Daolang Jujuli(J5),Xia Daolang Jujuli(J6),Luntai Jujuli(J7),Dong Kumuli(K2),and Shache Hongli(R1);Group Ⅲ,with moderate cold resistance,including 17 germplasms such as Dongli(D3),Aigenchak(Q13)and Kuche Jujuli(J4);Group I,exhibiting weak cold resistance,including the remaining 28 germplasms.The findings of this study could provide references for the future development and introduction of cold-resistant pear germplasm resources.
