Identification and Expression Analysis of Gibberellin 3-β-Dioxygenase Gene Family in Tea(Camellia sinensis)

Abstract:Gibberellins play critical roles in plant growth and development,and the gibberellin 3-beta-dioxygenase is a key enzyme in gibberellin synthesis.To investigate the role of GA3ox gene family in growth and development of tea shoots,in this study,four CsGA3ox gene family members were identified by homolo-gous comparison between the GA3ox family of Arabidopsis and the whole genome sequence of tea.Their physi-cochemical properties,subcellular localization,structure,evolutionary features,and the expression patterns at different stages of shoot development were analyzed.The results showed that the GA3oxs encoded 345 to 375 a-mino acids and all of them were hydrophilic proteins with no transmembrane structural domain.The structures of the proteins were mainly composed of α-helix and irregularly coiled,and contained the conserved structural domain of 2OG-Fe(Ⅱ)oxy.Phylogenetic tree analysis showed that the four CsGA3oxs were paralogous genes.The results of qRT-PCR analysis showed that the expression of CsGA3ox showed the decreasing trend during the growth of tea shoots,indicating that all the four CsGA3ox genes were involved in new shoot development of tea.The expression of GA3ox1 was the highest in the new shoots of tea and showed significant differences in different periods,which indicated that this gene was crucial for the growth of new shoots.
