Transcriptome Analysis of Pugionium dolabratum Seedlings under Drought Stress

Abstract:To explore the molecular mechanism of Pugionium dolabratum Maxim response to drought stress,explore potential drought resistance genes,and cultivate new varieties with drought resistance,tran-scriptome sequencing was carried out on the leaves of P.dolabratum under different drought stress levels.A to-tal of 538 differentially expressed genes were screened out by transcriptome sequencing,mainly in the path-ways of plant hormone signal transduction,plant pathogens,starch and sucrose metabolism,nitrogen metabo-lism,and RNA transport.Analysis of secondary metabolic pathways such as plant hormone signaling and ascor-bic acid metabolism showed that the expression levels of related genes were significantly increased in stressed leaves.The results of GO enrichment analysis showed that metabolic process,catalytic activity,antioxidant ac-tivity,protein-binding transcription factor activity and other biological pathways related to water stress,and molecular function and cell composition had significant changes.The leaf tolerance of P.dolabratum to drought stress was mainly related to the degree of gene response to stress,and P.dolabratum might adapt drought stress through signal transduction and secondary metabolism.The results of this study could provide references for further research of the molecular mechanism of P.dolabratum response to drought stress.
