Abstract:Protochlorophyllide oxidoreductase(POR)is an essential enzyme for chlorophyll synthesis, which plays the key role in chlorophyll metabolism. In this study, four CsPOR genes were identified based on the whole genome data of tea plant(Camellia sinensis),and the bioinformatics analysis and gene expression were carried out.The results showed that the CDS length of the four CsPOR genes was in the range of 1 200 and 1 368 bp,and the encoded proteins dominated by basic amino acids were stable and hydrophilic proteins. The sequence of CsPOR1,CsPOR2 and CsPOR3 proteins contained only cl21454 domain, while the se-quences of CsPOR4 contained cl21454 and cl47985 domains.CsPOR1,CsPOR2 and CsPOR3 had close ge-netic relationships.CsPOR proteins located in chloroplast, and had no transmembrane structure and signal peptide;their secondary structure was mainly composed of α-helix and random coil, and their tertiary structure was similar. The promoter of CsPOR genes contained many elements of light regulation, plant hormones and stress response.Furthermore, the expression of CsPORs in different tissues and tea plants with different leaf colors showed that the CsPORs mainly expressed in tea leaves, and the expression level affected the coloration of tea leaves. This study provided a theoretical basis for the further study of biological functions of CsPOR gene family.
