Effects of NaCl Stress on Physiological Indexes and Saponin Content of Adventitious Roots of Panax quinquefolium L.

Abstract:The purpose of this experiment was to investigate the effects of NaCl stress on physiological in-dexes and saponins content of adventitious roots of Panax quinquefolium L..In this experiment,the adventi-tious roots of P.quinquefolium were used as materials and four treatments of 0 mmol/L NaCl(CK),100 mmol/L NaCl(N1),200 mmol/L NaCl(N2)and 400 mmol/L NaCl(N4)were set,and the activity of antioxidant enzymes,content of osmotic regulatory substances,accumulation of total saponins,monomer saponins Rb1 and Re,and expression of HMGR,DS,D12H and P6H key enzyme genes in ginsenoside biosynthesis pathway were determined.The results showed that the proline and malondialdehyde(MDA)contents in adventitious roots of P.quinquefolium increased gradually with the increase of NaCl concentration and stress time.The ac-tivities of peroxidase(POD)and catalase(CAT)in adventitious roots of P.quinquefolium increased under N1 treatment and increased first and then decreased under N2 and N4 treatments,which were significantly higher compared with CK.Under N4 treatment for 48 hours,the total saponins content in adventitious roots of P.quinquefolium reached the maximum value,which was 28.42%more than that of CK.N2 and N4 treatments for 48 hours were conducive to the accumulation of Rb1 and Re monomonins in adventitious roots of P.quin-quefolium.The gene expression level of HMGR and D12H decreased firstly,then increased and then de-creased,while that of DS and P6H increased first and then decreased.The results of this study could provide theoretical bases for salt-resistant variety breeding and germplasm resource innovation of P.quinquefolium.
