Identification of Sorghum ABC Gene Family and Expression Analysis under Salt Stress

Abstract:ABC transporter(ATP-binding cassette transporter)family is one of the largest families of transporters in the plants,which can transport metabolites across the cell membranes and plays an important role in plant growth and development and resistance to stresses.In this study,the sorghum(Sorghum bicolor)ABC genes were identified in the whole genome,and 135 SbABC genes were screened out.According to the protein domain,they were divided into eight subfamilies,and named according to the chromosome position.Their evolutionary relationship,gene structure,protein motifs and promoter cis-acting elements were predicted and analyzed,and also the analysis of their expression patterns under salt stress.The results showed that these SbABC genes were unevenly distributed on ten chromosomes of sorghum,and the number of SbABC genes on chromosome 3 was the most.By constructing the phylogenetic tree,it was found that the clustering relationship of ABC genes in sorghum and Arabidopsis thaliana was consistent with the domain classification.Motif analysis showed that some motifs existed in most SbABC proteins,while some only existed in specific subgroups,which might be related to their functional differentiation.The promoter of SbABC genes contained the cis-acting ele-ments such as abscisic acid response,stress response and MYB recognition site elements,which were inferred to be related to sorghum growth and stress response.Through the analysis of transcriptome data,it was found that most of SbABC genes were expressed in roots,stems and leaves.Under the treatment of exogenous ABA(100 μmol/L)and NaCl(150 mmol/L),the expression of SbABCF3 and SbABCG22 genes were significantly induced.These results would be helpful to understand the role of ABC gene family in sorghum growth and salt tolerance,and provide new ideas for the research on sorghum salt tolerance mechanism and the cultivation of salt-tolerant crops.
