Effects of Magnaporthe oryzae Infection on Expression of Genes Related to Nitrogen Absorption and Utilization Pathways in Rice

Abstract:Rice blast is the most devastating fungal disease in rice production.High nitrogen fertilizer ap-plication tends to promote rice susceptibility to blast,however,the underlying genetic basis remains unclear.To elucidate the role of nitrogen uptake and utilization pathways during Magnaporthe oryzae infection,in this study,the expression patterns of genes associated with rice nitrogen absorption,metabolism and utilization sig-naling pathways were analyzed by real-time RT-qPCR after blast inoculation.The results showed that among the 33 genes regulating nitrogen uptake and utilization,22 genes including 8 nitrate transporter genes,3 am-monium transporter genes,3 nitrogen assimilation-related genes,5 nitrogen signaling regulatory genes,and 3 nitrogen utilization-related genes were significantly up-regulated at 4 hours after infected by a compatible M.oryzaerace,which showed no significant changes compared to the control during the middle to late infection sta-ges(8~96 h);28 genes including 11 nitrate transporter genes,4 ammonium transporter genes,5 nitrogen as-similation-related genes,5 nitrogen signaling regulatory genes,and 3 nitrogen utilization-related genes were down-regulated upon inoculation with an incompatible M.oryzae race(4~96 h).These findings demonstrated that the activation of nitrogen absorption,metabolism,and utilization signaling pathways played critical roles in facilitating early M.oryzae infection,which could provide references for further unraveling the molecular mechanisms of nitrogen-induced susceptibility to rice blast.
