Effects of Arginine on Morphological and Physiological Characteristics of Rice Seedlings under Low Temperature Stress

Abstract:This experiment used Oryza sativa'Xiangzaoxian 45'as test material and conducted under the artificially simulated low-temperature(4 ℃)environment.With spraying distilled water followed by nor-mal-temperature(25 ℃)treatment(CK1)and low-temperature(4 ℃)treatment(CK2)as control groups,spraying arginine solutions at different concentrations of 0.025,0.25,2.5 and 25 mmol/L(labeled as T1,T2,T3 and T4,respectively)were set to study their effects on the morphological and physiological character-istics of rice seedlings under low-temperature stress at 12,24 and 36 hours.The results demonstrated that low-temperature stress significantly inhibited plant height,aboveground fresh weight and chlorophyll biosynthesis in leaves.However,appropriate arginine concentrations effectively alleviated these inhibitions.After 12 hours low-temperature treatment,T2(0.25 mmol/L arginine)significantly increased plant height by 6.73%com-pared to CK2,and also increased the total root length,root surface area,root tip number and root branching number by 2.84%,10.06%,3.30%and 4.20%,respectively.After 24 hours low-temperature stress,T2 sig-nificantly enhanced plant height and stem diameter by 15.85%and 6.73%,respectively.After 36 hours of low-temperature treatment,T2 increased the aboveground fresh weight by 10.16%compared with CK2.After 12 hours and 24 hours low-temperature exposure,T2 significantly increased the chlorophyll b and total chlorophyll levels by 59.13%,31.74%and 94.43%,29.95%,respectively,compared to CK2.Arginine application en-hanced the peroxidase(POD)and catalase(CAT)activities in both leaves and roots under low-temperature stress.After 24 hours treatment,the activities of SOD and POD in leaves treated with T2 were significantly higher than those of CK2,with the increase rates as 29.69%and 30.53%,respectively.In sum,spraying 0.25 mmol/L arginine effectively alleviated the adverse effects of low-temperature stress on rice seedling morphologi-cal development and physiological activity,thus enhanced the cold resistance during seedling stage.
