Effects of Low Temperature Stress on Physiological and Biochemical Characteristics of Four Species of Bamboo and Comprehensive Evaluation on Their Cold Tolerance

Abstract:The experiment was conducted to compare the physiological changes in leaves of four bamboo species under low-temperature stress and assess their cold tolerance,in order to provide bases for introduction and cultivation of cold-resistant bamboos.Four temperature treatments(0,-5,-10 and-15 ℃)were set to study their effects on chlorophyll(Chl)content,relative conductivity(REC),antioxidant enzymes activities,soluble protein(SP)content,and malondialdehyde(MDA)content in leaves from the upper,middle and lower parts of four bamboo species(Phyllostachys bambusoides,Phyllostachys nigra,Bambusa ventricosa and Pleioblastus amarus).The results showed that as temperature decreased,the REC of leaves from different parts of the four bamboo species generally increased,the Chl content first rose then either fell or stabilized,the per-oxidase(POD)and catalase(CAT)activities displayed the increasing-decreasing-increasing trend,the su-peroxide dismutase(SOD)activity and SP content mostly showed the decreasing-increasing-decreasing pat-tern,and the MDA content initially decreased then increased.The cold tolerance of leaves varied across differ-ent parts of the plant.The upper leaves of Phyllostachys bambusoides,lower leaves of Phyllostachys nigra,and middle leaves of Bambusa ventricosa and Pleioblastus amarus had less cold tolerance compared with the other parts.Principal component analysis revealed that Chl content,SP content and POD activity had larger load values indicating significant effects on physiological changes in bamboos under low-temperature stress.Using membership function method,seven physiological and biochemical indexes of the four bamboo species were se-lected to comprehensively evaluate their cold tolerance,and the results indicated that their cold tolerance showed Phyllostachys bambusoides>Phyllostachys nigra>Bambusa ventricosa>Pleioblastus amarus.This conclu-sion could provide the theoretical basis for scientific cultivation and frost protection of leaves from different parts of these four bamboo species.
