Expression of Fiber-1 Protein of Duck Adenovirus Type 3 Based on a Novel Insect Baculovirus flashBACTM Expression System

Abstract:In recent years,duck adenovirus type 3(DAdV-3)has been reported in multiple regions,re-sulting in substantial losses to China's poultry industry.The fiber protein(Fiber)plays a critical role in media-ting DAdV-3 infection and inducing humoral immunity.In the present study,the Fiber-1 protein of DAdV-3 was recombinantly expressed using the insect baculovirus flashBACTM expression system to obtain the target protein.Owing to the integration of the LacZ gene into Autographa californica multiple nucleopolyhedrovirus(AcMNPV)in this system,LacZ expression was initiated post-recombination,enabling the screening of positive viruses via blue-white selection.This approach obviated the need for plaque screening,thereby considerably reducing the experimental time.Western blot analysis validated the successful expression of Fiber-1 protein in Sf9 insect cells.The results of this study could lay a foundation for further investigating the role of Fiber-1 protein in the infection mechanism of DAdV-3 and for the development of related biological products.
