Detection of Tomato brown rugose fruit virus and Screening of Resistant Sources

Abstract:In this study,the RT-PCR detection was carried out on suspected viral disease samples,and the amplified products were sequenced and the phylogenetic analysis was conducted,which verified that the samples were infected by Tomato brown rugose fruit virus(ToBRFV).The ToBRFV could be detected in seeds of infected tomato fruits,but could not be detected in seedings grown from the infected seeds,which indicated that ToBRFV was mainly existed in seed coat.The virus could be inactivated when the infected seeds were dis-infected with 10%of trisodium phosphate.Twenty-two Solanum pimpinellifolium resources were inoculated and identified by mechanical inoculation method at seedling stage,and two materials tolerant or resistant to To-BRFV were obtained,which could provide resources for resistant variety breeding.
