Identification and Expression Analysis of Glycoside Hydrolase GH3 Gene Family in Nicotiana tabacum L.

Abstract:The glycosyl hydrolase GH3 gene family plays significant roles in sugar metabolism and growth and development in plants.However,the GH3 gene family has not been identified and analyzed in tobacco.In this study,we employed bioinformatics methods to identify the GH3 gene family in the tobacco genome and in-vestigated the expression patterns of GH3 gene family members in different tissues and under stress conditions.The results showed that a total of 35 GH3 genes were identified in the tobacco K326 genome,and 23 of which were located on 14 chromosomes with 6 pairs of collinear genes.The GH3 family proteins were found to be heat-stable,and their upstream promoter regions contained multiple hormone and stress-responsive elements.Transcriptome data revealed that a majority of GH3 genes were highly expressed in the shoot apex of tobacco seedlings,while a few genes showed high expression in root and stem.Moreover,the expression of GH3 gene family members in tobacco were influenced by the duration of light exposure,and had different response to drought stress.qRT-PCR analysis of the selected four NtGH3 genes indicated that NtGH3-5,NtGH3-19,Nt-GH3-22 and NtGH3-26 were potentially involved in response to low temperature and ABA treatments.These findings could provide a basis for further understanding the GH3 gene family and their functions in tobacco.
