Effects of Different Concentrations of Phlorizin on Root Growth and Physiological Characteristics of Apple Rootstock

Abstract:In this experiment,the apple rootstock tissue-cultured seedlings were used as materials to study the effects of different concentrations(0,0.1,0.5,1.0 and 1.5 mmol/L)of phlorizin on the seedling growth,root system architecture,and root physiological and biochemical characteristics under sand culture conditions.The results showed that compared with the control(CK),the root growth of apple rootstock seed-lings was significantly inhibited by applying phlorizin at different concentrations.The fresh and dry weights of tissue-cultured seedlings decreased significantly;the root length,root surface area,root volume,average root diameter and net growth amount of plant height(except 0.1 mmol/L treatment)were significantly decreased with the increase of phlorizin concentration;the activities of superoxide dismutase(SOD),peroxidase(POD)and catalase(CAT)decreased,and the activities of chalcone synthase,chalcone isomerase and phlorizin hydrolase were also significantly reduced by exogenous phlorizin stress;the contents of MDA,superoxide anion and proline increased significantly,and the soluble protein content decreased significantly.In summary,with the increase of phlorizin concentration,the activities of antioxidant protective enzymes and soluble protein con-tent decreased,the contents of MDA,superoxide anion and proline increased,so the root growth of apple root-stock tissue-cultured seedlings was inhibited and then the growth of seedlings was inhibited.
