Abstract:Marking target microorganisms with green fluorescent protein (GFP) is an important means of studying the interaction between microorganisms and hosts. In this study, the shuttle vector pGFP4412 was introduced into the siderophore-producing bacterium Paenibacillus illinoisensis YZ29 by electroporation, and the YZ29-gfp strain successfully expressing GFP was obtained. The colonization of YZ29 on the peanut root surface and in the soil was studied by double-resistant plate screening combining with fluorescence microscope observation. The results showed that the green fluorescence could be observed under the blue light with excitation wavelength of 488 nm.The YZ29-gfp could colonize in the peanut rhizosphere and non-rhizo-sphere soils under pot culture.Under the condition of laboratory culture,it could colonize on the peanut root surface and could not colonize in the root. This showed that YZ29 could effectively colonize in the peanut rhizosphere, which layed a foundation for growth-promoting and biocontrol effects of peanut.
