Effects of Seed Priming on Germination of Platycodon grandiflorum Seeds and Seedling Growth under Salt Stress

Abstract:In order to explore seed germination characteristics and seedling growth of Platycodon grandiflorum under salt stress after seed priming,the priming experiments were conducted with NaCl,CaCl2 and GA,and the seed germination,seedling growth and physiological characteristics were compared between with seed priming and without priming.The results were as follows:the priming solutions of 1% NaCl,1% CaCl2,1 % NaCl + 1% CaCl2 and 1% NaCl + 1% CaCl2 + 750 mg/L GA could increase the seed germination rate and seedling biomass,in which,the priming effect was the best with 1% NaCl + 1% CaCl2 + 750 mg/L GA as priming solution,so it was selected as the optimum priming solution for Platycodon grandiflorum.Priming with 1% NaCl + 1% CaCl2 + 750 mg/L GA could strengthen the salt-resistant ability of Platycodon grandiflorum seeds,improve the seed germination characteristics and make the seedling emergence uniformly.Under the 25 mmol/L NaCl stress,the plant height,root length,and ratio of root to stem were higher than those of control (CK) whether seed primed or not,which implied that P.grandiflorum seeds could germinate and grow well under the salt stress of 25 mmol/L NaCl.Under the same NaCl stress,the plant height and root length of seedlings were significantly higher with seed priming than those without seed priming.In conclusion,the P.grandiflorum seeds could endure 25 mmol/L NaCl stress,and priming with 1% NaCl + 1% CaCl2 + 750 mg/L GA could significantly increase the seed germination characteristics and seedling salt tolerance.
