Clone and Expression Analysis of Wild Peanut Gene AdWRKY37

Abstract:Peanut (Arachis hypogaea L.) is one of the most important economic crops in China.Wild peanuts have many good traits which are lacking in cultivated peanut varieties.Wild peanuts are indispensable germplasm resources as well as genetic resources for improving cultivated peanut varieties.It has been demonstrated that WRKY family transcription factors are involved in various disease resistance in plant.In our study, AdWRKY37, which belongs to WRKY family, was cloned and identified from wild peanut.Furthermore, bioinformatics approach was performed to analyze its gene and protein sequences.The results showed that AdWRKY37 protein contained NBS-LRR domain.Besides, salicylic acid(SA)-and jasmonic acid (JA)-induced elements were also identified in the promoter sequence of AdWRKY37.The expression analysis using qRT-PCR method revealed that the gene expression amount of AdWRKY37 obviously increased under the induction of Ralstonia solanacearum as well as SA and JA treatments.All the results indicated that AdWRKY37 in wild peanuts might participate in resisting to Ralstonia solanacearum.
