Breeding of Radish Recessive Nuclear Male Sterile Line 136A and Its Fertility Study

Abstract:136A,a new type of radish nuclear male sterile material,was found from the natural popula-tion.Through crossing with fertile plants in 178 hybrid combinations,136B was selected,which had the abili-ty to maintain the sterility of 50%.Then the stable and homozygous male sterile line 136AB was bred through several generations.The field genetic experiment showed that the sterile trait of 136A was stable,and ex-pressed the nuclear sterile genetic feature which was controlled by a single recessive gene.The cytological ob-servation indicated that the pollen of 136A was thoroughly abortive,the pollen development began abnormal from the tetrad stage,and pollen abortion was mainly induced by the rupture of pollen wall.The PCR results showed that 136A did not contain orf138 gene,which was the major gene of Ogura male sterile type.The mi-tochondrial and chloroplast DNA sequencing results also showed that 136A had the normal fertile cytoplasm.
