Optimization of Preparation Technology for Special Phosphate Solubilizing Microorganism Agent in Brown Soil Tea Garden by Response Surface Methodology

Abstract:The response surface methodology was used to optimize the preparation technology for special phosphate solubilizing microorganism agent in brown soil tea garden.Three key factors that affected the prepa-ration technology were screened out by Plackett -Burman design,including the ratio of water to carrier (X1 )、fermentation temperature (X4 )and drying temperature (X7 ).A quadratic regression equation model was es-tablished according to the steepest ascent experiment,central composite design and response surface methodol-ogy with the number of effective living bacteria in agent as response value.The maximum number of effective living bacteria predicted by this model was 5.01 ×10 10 per gram and the optimal preparation conditions were as follows:the ratio of water to carrier as 1.8,the amount of bacterial inoculation to carrier as 12.5% and the fermentation and drying temperature as 30℃ and 41℃ respectively.Under the optimization process condi-tions,the number of effective living bacteria in agent reached 5.19 ×10 10 per gram,which was basically con-sistent with the predicted maximum value.These results suggested that the prediction model was reliable.
